BMP-4 is a TGF-beta superfamily ligand that is widely expressed from early embryogenesis through adulthood. It plays an important role in mesenchyme formation, epidermal determination, suppression of neural induction, the development of multiple organs, and tissue repair (1-5). The human BMP-4 precursor contains a 273 amino acid (aa) propeptide and a 116 aa mature protein (6). Processing of the propeptide by furin or proprotein convertase 6 enables the formation of the mature disulfide-linked homodimeric BMP-4 and facilitates its secretion. Similar intracellular processes may lead to the formation and recreation of BMP4/BMP7
disulfide-linked heterodimer (7-9). Mature human and mouse BMP-4 share 98% aa sequence identity. Human BMP-4 shares 85% aa sequence identity with human BMP-2 and less than 50% with other human BMPs. Compared to BMP-4 homodimers, BMP-4/BMP-7 heterodimers exhibit a greater potency in inducing osteogenic differentiation (9). In Xenopus, the heterodimers can also induce the formation of mesoderm, whereas BMP-4 homodimers only provide ventralizing signals for existing mesoderm (10). BMP-4 signals through tetrameric complexes composed of type I (primarily Activin RIA or BMPR-IA) and type II (primarily Activin RIIA or BMPR-II) receptors (11, 12). The bioavailability of BMP-4 is regulated by its interaction with multiple proteins and glycosaminoglycans (13-15).

CF stands for Carrier Free (CF). We typically add Bovine Serum Albumin (BSA) as a carrier protein to our recombinant proteins. Adding a carrier protein enhances protein stability, increases shelf-life, and allows the recombinant protein to be stored at a more dilute concentration. The carrier free version does not contain BSA.

What formulation is right for me?
In general, we advise purchasing the recombinant protein with BSA for use in cell or tissue culture, or as an ELISA standard. In contrast, the carrier free protein is recommended for applications, in which the presence of BSA could interfere.

尊敬的客户您好:

很高兴您选择优宁维进行科研服务,为了保障我们双方的共同利益,精诚良好的合作,避免产生不必要的责任纠纷,请您认真阅读以下内容并配合我们事宜,若您同意以下内容,请点击同意,进入网站下单界面。

1.为保证数据质量,您需严格按要求准备样本和试剂:

1)血清、血浆、尿液、灌洗液、组织、细胞沉淀、裂解蛋白等,未经过反复冻融,且用量满足实验要求的最低标准。

2)组化实验组织样本,需经过10%中性缓冲福尔马林固定24-72小时后,转移到75%酒精中送样,且保证样品管上标记清晰,不会被酒精渗漏模糊掉字迹。

3)组化实验蜡块和切片样本,常温运输,尽可能使用盒子装蜡块,避免互相之前摩擦影响样本;切片使用切片盒放置,避免玻片之间相关刮创。

4)您必须明确说明提供给乙方的实验材料的安全性,确保无致病性、无传染性,对人、动植物和环境友好安全,以保证实验技术人员的安全。

5)珍贵样本,请您只提供满足实验服务合同的所需量,剩余样本自己保存好以免造成样本的浪费,耽误您的实验进展。

6)甲方任何样本准备过程中的疑问,均可以咨询乙方。或者发邮件至labex@univ-bio.com,或者拨打4008-168-068转3。

2.为保证实验准确性,您需知悉以下内容:

1)非试剂盒验证样本,存在检出率低的风险,客户知晓并同意的情况下,Labex配合开展实验。

2)检出率是在MSD标准曲线范围内的数据,样本内因子含量超出标准品检测范围的情况下:若低于MSD标准曲线范围,则无法检出;若超过标准品检测上限,需实进一步稀释样本优化实验。MSD技术是目前市场上检测范围最宽(横跨4-5log)的多因子检测技术,但蛋白质组几万种蛋白浓度横跨10 log,不能一概而论。

3.实验服务过程中样本和试剂运输要求:

1)您可自行送样(方式包括快递以及亲送)到我们指定地点,但过程中要保证正确的运输条件,干冰货物或冰袋运输务必保证干冰充足和冰袋低温。并及时通知到我们,方便我们接收。

您还需要准备一份随样单(合同附件所示),标注您的姓名、合同号、样本信息、试剂信息,随快递一起寄送。

若由于您运输条件的原因,导致实验结果问题,相关责任由您承担。

2)若您需要我们协助寄送样本时,我们会收取合理的寄送费用,以我们标准合同上所列收费为准。

4.实验服务合同结束后样本和试剂处置须知:

请您下单前确定实验服务合同结束后,样本处置情况。我们将严格按照合同约定执行。若您选择实验结束后样本,不保留,丢弃,我们会在实验结束后按国家标准自行处置掉相关实验材料。若您需要回寄相关样本,在您支付一定的运输费用后,我们将剩余样本在规定时间内返还给您。若您选择到付,需要保证您的样本是常温和蓝冰运输的,更高要求的运输条件不支持货到付款。若您不确定是否回寄,我们将保持您的样本1个月的时间,如超期,样本我们将自行处置,不在另行通知您。 如果您选择我们回寄样本给您,我们将快递信息邮件告知您,由您跟踪物流信息,并及时确认。如因您个人原因或者第三方物流原因造成的样本邮寄过程中的损耗,我们不承担任何责任。

5.实验服务种实验服务数据:

1) 我们承诺按照《实验样本信息及要求》中的实验方案进行实验并提供客观实验结果,但由于科研实验中存在诸多不确定因素以及未知难度和风险而导致结果的不确定性,不能保证所有的实验进度和实验结果完全达到甲方预期。在排除实验操作问题导致不良结果的前提下,您应尊重客观事实,承担实验结果与预期不一致的结果,包括实验结果是阴性的风险。因此在得不到预期结果的情况下,您仍应支付本次实验服务的全部已发生费用。

2) 实验服务是由专业技术达标的实验人员,遵循科学实验方法和标准操作流程,依照服务合同完成,优宁维保证将客观真实科学严谨的实验结果给到客户,但单次实验不足以阐明某些复杂的科学问题。

3) 您在收到实验数据后,5个工作日内(荧光相关实验3个工作日内)对实验数据质量提质疑,我们有义务解答。若是我们实验过程中的问题,我们承担全部责任;若是您验设计问题,您承担全部责任。我们承诺1个月的数据复核期,欢迎后续咨询。

6.客户知识产权保密协议:

1) 实验服务过程中产生的实验数据的知识产权归您所有,我们承诺不得以任何形式向除甲方以外的第三方提供该实验数据。对于相关的实验技术所有权归我们所有,您不得主张权利。

2) 我们双方共同致力于维护对方的商业机密和技术机密,包括本次合同所签订的文件,您都有义务进行保密。

7.服务承诺

如由于实验客观性需要重复实验、自然灾害、政府行为或其他乙方无法预见、无法避免和无法克服等不可抗力的客观原因造成合同无法按期履行的,我们将及时通知甲方,双方将视情况决定是否继续履行合同。如果由于客观原因造成某些实验项目不能执行的,合同结束时,退回相应款项。
